The EcoPro™ T7 System represents a significant improvement in coupled in vitro transcription/translation. The highly optimized EcoPro T7 Extracts efficiently synthesize full-length proteins from genes downstream of E. coli T7 promoters using circular or linear DNA templates, including PCR amplified DNA. The EcoPro extracts are prepared with a proprietary fractionation process* that produces higher protein expression levels and predominantly full-length protein, with a marked reduction in the secondary products that are commonly observed with other prokaryotic in vitro expression systems.

The EcoPro T7 Extract is supplemented with T7 RNA polymerase, such that protein synthesis is directed from T7 promoters in addition to E. coli promoters. Optimal translation is achieved using templates that carry a strong prokaryotic ribosome binding site (RBS; e.g., AAGGAG) 6 to 11 base pairs upstream from the ATG initiation codon. The EcoPro T7 Extract contains optimal levels of all components needed for template-dependent transcription and translation, reducing the time and expense of reaction assembly. The EcoPro T7 System can be easily integrated with a variety of non-isotopic detection methods including the FRETWorks™ S•Tag™ Assay.
