A naturally-occurring antagonist to angiopoietin-1. Closely related to the pro-angiogenic factor Ang-1 and binds with similar affinity to the Tie-2/Fc receptor tyrosine kinase. Competitively inhibits Ang-1-induced endothelial cell responses mediated by Tie-2/Fc. Disrupts vasculogenesis in developing embryos by antagonizing Ang-1.

Activity
0.5-100 ng/ml is the linear range of binding of angiopoietin-2 to Tie-2/Fc. Assay was performed with immobilized Tie-2/Fc (4 µg/ml).
Biological activity
Linear range of biotinylated angiopoietin-2 binding = 0.5-100 ng/ml based on its ability to bind immobilized Tie-2/Fc (plated at ~400 ng/well) in a functional ELISA.

Source
cDNA sequence corresponding to human angiopoietin-2 was expressed with a C-terminal 6X histidine tag in a mouse myeloma cell line, NSO, and then biotinylated.
