   Varying concentrations of enzyme were mixed with 1 µM substrate in cAB containing 75 µM ATP. Relative fluorescence units (RFU) decrease as a function of enzyme activity.  Varying amounts of ATP were used in the absence or presence of enzyme. Percent activity was calculated based on the ΔRFU between the samples.  Varying amounts of Wortmannin in 0.05% DMSO were added to enzyme (32 nM) and substrate (1 µM) in cAB containing 20 µM ATP.  Statistical data were produced using 0% (top) and 100% phospho-calibrators (bottom) or 27 nM PI 3-Kα (middle). Z′-factors of 0.8 and 0.7 were obtained using phospho-calibrators or reacted samples, respectively. A Z′-factor of >0.5 indicates a robust assay. |
