This kit allows the quantitation of apoptotic cells in vitro by DNA affinity-mediated capture of free nucleosomes followed by their anti-histone-facilitated detection. In this assay, mono- and oligonucleosomes are captured on pre-coated DNA binding proteins. Anti-Histone 3 (H3) biotin-labeled antibody then binds to the histone component of captured nucleosomes and is detected following incubation with streptavidin-HRP conjugate. HRP catalyzes the conversion of colorless tetramethylbenzidine to blue. The addition of stop solution changes the color to yellow, the intensity of which is proportional to the number of nucleosomes in the sample. By comparing the absorbance of unknown with that obtained from the standards, one can assign a nucleosome unit value to the unknown sample. Can be used to assign apoptotic index.
