LiChrospher® SorbentsSorbents for High-Performance Preparative ChromatographyLiChrospher for preparative high-performance chromatography is a spherical and porous silica gel material characterized by:
Underivatized silica gels still play an important role in preparative chromatography. For routine applications, it is important that spherical silica gels are produced according to the same stringent guidelines as irregular Merck KGaA silica gels. This means that only those spherical silica gels are released that meet all required specifications with respect to pore structure, specific pore volume, specific surface area and chemical purity as well as complying with additional important chromatographic parameters. Only by guaranteeing such parameter is it possible to produce products for economic and efficient use. Independent of the chromatography process used to develop a particular method (DC, TLC, HPTLC, HPLC, preparative column chromatography), its direct and rapid transfer to production scale is guaranteed. Identical raw materials used in spherical silica gel manufacturing guarantees direct scale-up without risk or lost time. LiChrospher® RP MaterialGuarantee for Selectivity and ReproducibilityMany chromatographic separations can be carried out more easily, more rapidly and more economically by using RP-sorbents. Merck KGaA supplies the most important RP-modifications in the form of LiChrospher 100 RP-18, LiChrospher® WP 300 RP-18 and LiChrosphe 60 RP-select B in two different particle sizes (12µm and 15µm): LiChrospher 100 RP-18 is characterised by its high degree of surface coverage and hence extraordinarily high lipophilic behaviour. LiChrospher 60 RP-select B has in addition, specific matrix properties, which enable the separation of components with basic properties under reversed phase conditions while minimizing/eliminating tailing effects. LiChrospher WP 300 RP-18 based on a wide-pore LiChrospher of 300Å pore size, is particularly suitable for the separation of peptides, low-molecular weight proteins and tRNA molecules. One of the most important characteristics in chromatography is selectivity. Only stationary phases demonstrating identical batch-to-batch selectivity can guarantee reliable reproducibility of the chromatographic separation and hence high yield when used in the preparative mode. In order to guarantee such batch-to-batch reproducibility over a period of years, LiChrospher 100 RP-18 and LiChrospher 60 RP-select B have been subjected to comprehensive selectivity documentation; in addition, a corresponding selectivity test is carried out prior to each batch being released. Using specific, chemically different test solutes under precisely defined, chromatographic conditions, this selectivity test enables the smallest changes in selectivity properties of the sorbents to be monitored. Thus, Merck can guarantee that LiChrospher 100 RP-18 and LiChrospher 60 RP-Select B deliver reproducible results within their specified narrow limits. For the user, this means the highest possible degree of reliability when using chromatographic separation techniques and purification techniques in the laboratory, pilot plant and production. The Advantages: Uniform selectivity for analytical and preparative materials enables reliable and rapid scale-up. Uniform batch-to-batch selectivity is at the same time a guarantee of reproducibility - over a period of years. Quality guarantees from Merck KGaA have been the standard for guaranteed quality in analysis for over 100 years. LiChrospher 100 RP-18 and LiChrospher RP-select B provide the guarantee of reproducibility in high performance preparative chromatography. |
